LC-MS Method Development for Simultaneous Analysis of Methamphetamine and its Main Metabolite Amphetamine

More than 230 million people worldwide have admitted to using an illicit drug at least once in their life time according to the World Health Organization. Methamphetamine is the second most commonly abused drug behind heroin. It is abused by more than 24.7 million people globally. It is also the most commonly abused drug among young adults of ages 18-25 in the United States, (Office of Applied Studies). This emphasizes the importance of the determination of methamphetamine and its metabolites. In our proposed research a sensitive LC-MS method will be developed for the simultaneous determination of methamphetamine and its main metabolite amphetamine. Chromatographic separation will be carried out using HPLC with C18 as the stationary phase and composition of acetonitrile and water containing formic acid as mobile phase. HPLC will be coupled with an ion trap mass spectrometer equipped with ESI technique for the detection of the eluted compounds. Rat serum will be spiked with methamphetamine and amphetamine and extraction techniques like SPE and dSPE will be used. After the successful extraction of analytes from serum sample, further analysis will be carried out using LC-MS. The method developed will be validated for linearity, accuracy, precision, LOD, LOQ, extraction recovery and specificity.
