Formulation, Optimization, and Evaluation of Oral Dissolving Film Strips of Verapamil Hydrochloride

Orally dissolving film strips (ODFS) are novel dosage forms that disintegrate and dissolve instantaneously when placed on the tongue or in the buccal cavity. This dosage form not only increases patient compliance of medications, but also improves bioavailability of the drug. The aim of this study is to formulate verapamil HCl ODFS with dissolution time less than 30 seconds, and also to study the film forming properties of two polymers hydroxypropyl methyl cellulose (HPMC) and LYCOAT® (Hydroxypropyl Pea starch). Two ODFS formulations containing verapamil HCl are prepared by solvent casting method using two different water soluble polymers, HPMC of E5, E15 grades and LYCOAT® of RS720, RS 780 grades. In this study, glycerin is used as plasticizer, maltodextrin as a film modifier and stevia as a sweetener. The prepared films are evaluated for thickness, weight variation, pH, folding endurance, uniformity in drug content and dissolution time.
