Title: Comparison of sfGFP expression in Geobacillus kaustophilus HTA426 transformed with plasmid pJZ04e vs. integrative plasmid pTM33c

The green fluorescent protein (GFP) is an established marker of cell expression, which has wide applications in molecular biology. In this study, a superfolder variant of GFP (sfGFP) will be cloned into two separate plasmids, pTM33c and pJZ04e, and inserted into a thermophilic bacterium, Geobacillus kaustophilus HTA426. The plasmid pTM33c is expected to integrate into the genome of G. kaustophilus and the plasmid pJZ04e is expected to multiply inside G. kaustophilus. The expression of sfGFP in G. kaustophilus HTA426 transformed by both the plasmids will be measured by a fluorescence assay and compared.
