Title: Role of NQO1 and Mitochondria in the Redox Activation of Quinoid Anticancer Agent in Human Lung Carcinoma and Colorectal Carcinoma Cells

In a previous study, we characterized two human cancer cell lines according to their activity and expression of NQO1 and found human lung carcinoma (A549) cells have high activity and protein expression of NQO1 compared to human colorectal carcinoma (Caco-2) cells. The rate of oxygen consumption and levels of ROS generated in both cell lines were studied with the help of oxygen polarograph, luminometer, and EPR to establish the involvement of NQO1 or mitochondria in redox activation of beta-Lp. It was observed that redox activation of beta-Lp was mostly mediated by NQO1 in A549 cells and in Caco-2 cells, mitochondria caused redox activation of beta-Lp. Further, to understand the mechanism involved in mitochondria-mediated redox activation of beta-Lp, we used different complex inhibitors like rotenone, myxothiazol, and antimycin and observed their effect on the production of ROS. With preliminary results, mitochondrial complex 3 seems to be the major site of redox activation of beta-Lp.
